I want to buy applications for the iPhone with an iTunes gift card, but I want to ensure you can before buying one.

If you are in the U.S.: Yes, if you're in Canada: No. If you are in Mexico: not even can buy songs on iTunes. Canadian law prohibits Apple from being able to use gift cards to pay for software. However, in the Apple site say Gift cards are good for all purchases from iTunes Store and are not aware of the inability to do with the App Store, if you live in Canada.
